I. 本实验室长期以来主要以细胞生物学、分子生物学和生物化学等为研究手段,在分子水平、细胞和亚细胞水平以及组织和个体水平上,从事干细胞增殖与分化、细胞周期调控、细胞核结构和动态变化及功能、Ran GTP酶功能调控等方面的研究。实验材料主要包括:培养细胞,小鼠,非洲爪蟾,果蝇等。
II. 已取得如下主要成绩:
1,发现Ran GTPase 控制核膜装配(Science, 2000; Curr Biol, 2011; J Cell Sc, 2012)。
2,发现Ran GTPase 控制纺锤体组装(J Cell Sci, 1999)。
3,建立了应用相关蛋白质小球和细胞(包括卵细胞)提取物为材料的非细胞体系核膜重建模式(Curr Biol, 2001, 2002; Eur J Cell Biol,2002)。
4,发现核膜物质运输相关蛋白importin alpha和beta 参与核膜装配(Curr Biol, 2002;Cell Res,2012)。
5,发现Ran 调控核孔复合体装配(Eur J Cell Biol, 2002;
Cell Res,2012)。
6,发现RCC1在染色体上定位所产生的Ran-GTP梯度是纺锤体正确装配所需要的(Curr Biol, 2002)。
7,发现Ran GTPase 通过调控核膜物质运输进而调控DNA 复制(J Cell Sci,1998)。
8,以实验证明细胞核纤层(Nuclear lamina)对细胞核结构的维持和DNA 复制起重要调控作用(J Cell Sci,1996)。
9,发现Nucleoplasin在非洲爪蟾卵提取物细胞凋亡过程中调控染色质的凝集(PNAS,2005)。
10,发现Aurora A/B, Plk1, Cdk1等激酶综合调控纺锤体装配(PNAS,2009,2013;Cell Res, 2008, 2011; J Cell Sci, 2009, 2013a, 2013b)。
11,发现CDK1, Aurora A/B和TPX2等综合调控纺锤体装配的新机制(J Cell Biol, 2015a; J Cell Sci, 2015;J Biol Chem, 2015)。
12, 发现去泛素化酶Ups16和Plk1综合调控动粒与纺锤体微管的结合及纺锤体装配的调控机制(J Cell Biol,2015b;CMLS,2017)。
13,发现细胞周期调控因子调控初级纤毛动态变化及其细胞信号转导功能(J Cell Sci, 2013, 2014;PLoS Biol, 2015; J Biol Chem, 2017a)。
14,发现DNA复制起始调控因子CDC6调控rDNA转录的新机制(J Cell SCI,2016)和调控中心体复制的新机制(NatureCommunications,2017)。
15,发现CDK4-Cyclin D1激酶调控细胞周期起始的新机制(J Biol Chem,2017b)。
